Cultivar ¢¥Blue Boy¢¥ of Standard Line and cultivar ¢¥California¢¥ of Optimara Line were experimented to elucidate optional concentration of either BA, NAA or 2,4-D for in vitro culture by modifying widely used MS cultural medium. The results are presented to be as follows:
1. Levels and ratio of BA and NAA were regarded to be the best combined and showed the most pronounced formation of calli and differentiation of adventitous shoots, if BA is maintained at 1§·/liter and NAA at 0.25§·/liter having 4 to 1 ratio,
2. Adventitous shoots and roots were also produced even when either BA or NAA alone are separately added.
3. Cultural medium containing 1 to 1 ratio of 2§·/liter BA and 2§·/liter NAA showed no adventitous shoot formation, and this is probably due to the fact that the high level of auxin suppresses the influence of cytokinin.
4. When BA and 2, 4-D were combined in vitro cultural medium, the best growth of plantlets were obtained with 2§·/liter BA plus 0.005§·/liter 2, 9-D, yet a rather compact appearance of plantlet formation was noticed if a medium of BA with the similar level of N:1A was used.
